Cyclic Nucleotide-Regulated Ion Channels: From Light Sensing to Pace Keeping

ثبت نشده
چکیده

The cyclic nucleotide-regulated ion channels are a heterogeneous superfamily of different ion channels that share common membrane topology and pore structure and display a cyclic nucleotide-binding domain in their C-terminal region. Members of this superfamily include cyclic nucleotide-gated (CNG) cation channels, hyperpolarizationactivated cyclic nucleotide-gated (HCN) cation channels and also several members of the voltage-dependent K channels such as the K V 10 (eag) and the K V 11 (erg) subfamilies. Channel regulation by cyclic nucleotides has been largely studied in the context of the CNG and HCN groups and will be the subject of this brief review. Noemí Bronstein-Sitton, Ph.D.

CryoEM structure of a prokaryotic cyclic nucleotide-gated ion channel.

Cyclic nucleotide-gated (CNG) and hyperpolarization-activated cyclic nucleotide-regulated (HCN) ion channels play crucial physiological roles in phototransduction, olfaction, and cardiac pace making. These channels are characterized by the presence of a carboxyl-terminal cyclic nucleotide-binding domain (CNBD) that connects to the channel pore via a C-linker domain. Although cyclic nucleotide b...

Modification of Cyclic Nucleotide–Gated Ion Channels by Ultraviolet Light

We irradiated cyclic nucleotide-gated ion channels in situ with ultraviolet light to probe the role of aromatic residues in ion channel function. UV light reduced the current through excised membrane patches from Xenopus oocytes expressing the alpha subunit of bovine retinal cyclic nucleotide-gated channels irreversibly, a result consistent with permanent covalent modification of channel amino ...

An inward current induced by a putative cyclic nucleotide-gated channel in rat cerebellar Purkinje neurons

The roles of cyclic nucleotide-gated (CNG) channels in sensory transduction have long been recognized. More recent studies found that CNG channels are distributed in multiple brain regions involved in memory and learning, including the cortex, hippocampus and cerebellum. These findings suggest that their functions are not limited to sensory perception, but also to neuronal plasticity phenomena,...

C-Terminal β9-Strand of the Cyclic Nucleotide-Binding Homology Domain Stabilizes Activated States of Kv11.1 Channels

Kv11.1 potassium channels are important for regulation of the normal rhythm of the heartbeat. Reduced activity of Kv11.1 channels causes long QT syndrome type 2, a disorder that increases the risk of cardiac arrhythmias and sudden cardiac arrest. Kv11.1 channels are members of the KCNH subfamily of voltage-gated K(+) channels. However, they also share many similarities with the cyclic nucleotid...
